Spotlight Issue on Signaling in Cardiac Metabolism: Transcriptional Control of Mitochondrial Biogenesis. the Central Role of Pgc-1α

چکیده
Although the concept of energy starvation in the failing heart was proposed decades ago, still very little is known about the origin of energetic failure. Recent advances in molecular biology have started to elucidate the transcriptional events governing mitochondrial biogenesis. In particular a great step forwards was taken with the discovery that peroxisome proliferator-activated receptor gamma coactivator (PGC-1α) is the master regulator of mitochondrial biogenesis. The molecular mechanisms underlying the downregulation of PGC-1a and the consequent decrease in mitochondrial function in heart failure are, however, still poorly understood. Indeed, the main pathways involved in mitochondrial biogenesis are thought to be uprather than down-regulated in pathological hypertrophy and heart failure. The current review summarizes recent advances in this field and is restricted to the heart when cardiac data is available. I. MITOCHONDRIAL BIOGENESIS:

Introduction: Death from cardiovascular diseases is one of the leading causes of death in the elderly; Although the role of exercise and herbs in heart health has been reported, their cellular-molecular mechanisms are not yet well understood. Therefore, the aim of the present study was to investigate the effect of eight weeks of continuous exercise with moderate intensity and intense periodicit...

Background: Type 2 diabetic patients have an abnormally high rate of mortality due to cardiovascular diseases. Given the adverse impact of diabetes on in heart cells and the role of exercise on mitochondrial biogenesis signaling, this study investigated the effect of eight weeks of aerobic exercise on PGC-1a and SIRT-1 gene expression in the myocardium of diabetic male Wistar rats. Materials a...
متن کاملThe Effect of Moderate and High Intensity Resistance Training on the Expression of PGC-1α, TFAM and AMPK of Cardiomyocytes in Elderly Rats
Background & objectives: Age-related cardiovascular dysfunction is often accompanied by impaired mitochondrial biogenesis and function. Exercise training can improve mitochondrial function and content in muscle to meet the energy demands of the cells. The purpose of the present study was to investigate the effect of moderate-intensity (MRT) and high-intensity (HRT) resistance training on the ex...
متن کاملThe PPARα-PGC-1α Axis Controls Cardiac Energy Metabolism in Healthy and Diseased Myocardium
The mammalian myocardium is an omnivorous organ that relies on multiple substrates in order to fulfill its tremendous energy demands. Cardiac energy metabolism preference is regulated at several critical points, including at the level of gene transcription. Emerging evidence indicates that the nuclear receptor PPARalpha and its cardiac-enriched coactivator protein, PGC-1alpha, play important ro...
متن کاملTHE EFFECT OF ENDURANCE EXERCISE ON THE CONTENT OF AMPK AND PGC-1Α PROTEINS IN THE LEFT VENTRICULAR HEART TISSUE OF RATS WITH TYPE 2 DIABETES
Background: One of the most important biological pathways involved in maintaining energy homeostasis is the AMPK PGC-1α pathway. Activation of this pathway through exercise can be important in regulating mitochondrial biogenesis processes and maintaining energy balance in diabetics. Therefore, the aim of this study was to investigate the effect of endurance exercise on the content of AMPK and P...
